Cleghorn graduated MD from the University of Edinburgh in 1783 and became a member of the Faculty of Physicians and Surgeons of Glasgow in 1786. He became the Faculty's President, 1788-1791 and then its librarian. In 1786 he was appointed the physician to the Town's Hospital. He was a lecturer at the University of Glasgow in Materia Medica, 1788-1791, and in Chemistry, 1791-1818. He was one of the first two visiting physicians at Glasgow Royal Infirmary, serving there for five years between 1795 and 1810, and was also one of the first men elected to the Royal's Board of Management.
