The house is famous as the birthplace of the anatomist William Hunter (1718-1783) and his brother John (1728-1793), two of ten children. William and John became estranged in 1780. William inherited the house c 1751, but when he died in 1783 he left the house to his nephew Matthew Baillie, rather than to John. However, Matthew transferred the house to John, and did not take up his inheritance until his uncle's death in 1793.
Matthew Baillie was also a noted anatomist and physician. His sister Joanna Baillie (1762-1851), the poet and playwright, lived at Long Calderwood for six years.
In 2004, the house is occupied by the Hunter House Museum
